ABSTRACT

Objective: Diabetic nephropathy is one of the most common microvascular complications of diabetes mellitus that finally leads to complete loss of kidney function. Therefore, this study aimed to evaluate the effect of crocin and losartan on TGF-ß gene expression and histopathology of kidney tissue in a rat model of diabetic nephropathy. Materials and Methods: Forty male Wistar rats were randomly divided into five groups (n=8): Untreated control, Diabetic (D), D + crocin, D + losartan, and D + losartan + crocin. Induction of diabetes was performed using streptozotocin (50 mg/kg/ Intraperitoneal injection). At the end of the eight-week period, the rats were sacrificed. Spectrophotometry measured serum glucose, urea, creatinine, and uric acid levels. Microalbumin and creatinine levels were measured in 24-hour urine. Real-time PCR was used to determine the relative expression of the TGF-ß gene in kidney tissue. Renal tissue histopathology was also examined. Results: The results showed that hyperglycemia increased biochemical factors associated with diabetes, TGF-ß gene expression, and kidney damage. Separate treatment with crocin and losartan led to a decrease in renal function factors and TGF-ß gene expression and improved kidney damage. Conclusion: Our results showed that crocin could improve kidney function in diabetic conditions. In addition, we showed that crocin increases the effectiveness of losartan. Consequently, we suggest that crocin in combination with chemical drugs can be a potential therapeutic agent for diabetes and its complications. Nonetheless, human studies are needed to make firm findings.

ABSTRACT

This study aimed to evaluate the effect of saffron, crocin, and safranal on serum levels of oxidants and antioxidants in diabetic rats. The authors searched the databases with standard keywords until June 8, 2021. The random-effects model was used to pool standardized mean differences (SMD) with 95% confidence intervals to assess the effects of saffron and its active component. To investigate heterogeneity, subgroup analysis and meta-regression were utilized. Begg and Egger's tests were used to measure publication bias. Our results showed that saffron, crocin, and safranal were able to significantly reduce the serum levels of oxidants with strong efficacy so that saffron had the highest effectiveness on serum malondialdehyde (SMD, -2.84 (µmol/L) [95% confidence interval (CI), -4.32 to -1.36]; p < .001, I 2 = 83.5%). In addition, saffron and its effective compounds were highly effective by increasing the serum level of antioxidants. In addition, saffron and its effective compounds were able to significantly increase the serum level of antioxidants with strong efficacy, while saffron had the highest effect on the serum level of total antioxidant capacity (SMD, 3.90 (µmol/L) [95% CI, 0.78-7.03]; p = .014, I 2 = 86.9%). The findings of this study show that treatment with saffron, crocin, and safranal by strengthening the antioxidant defense system and modulating oxidative stress shows antidiabetic effects in the diabetic model of rats, also these findings support the potential effect of saffron and its effective compounds for the management of diabetes and its complications. However, more human studies are needed.

ABSTRACT

BACKGROUND: Diabetes is a multifactorial and growing disease, one of the severe complications of which is diabetic nephropathy (DN), which is the most common cause of chronic renal failure. FERM domain containing 3 (FRMD3) is responsible for maintaining the shape and integrity of nephron cells, and bone morphogenetic protein 7 (BMP7) helps maintain function and reduce kidney damage. This study aimed to evaluate the effect of crocin and losartan on biochemical parameters and the expression of FRMD3 and BMP7 genes in streptozotocin (STZ)-induced diabetic rats. METHODS: Forty male Wistar rats were randomly divided into five experimental groups as healthy, diabetic control (D), crocin, losartan, and diabetic rats treated with losartan-crocin (n = 8). A single dose of STZ (50 mg/kg intraperitoneally injection) was used to induce diabetes. Four weeks after induction of diabetes, rats received crocin (50 mg/kg) and losartan (25 mg/kg) daily for four weeks orally. Rats were sacrificed at the end of the intervention, and blood samples were taken to determine serum levels of glucose, urea, creatinine (Cr), malondialdehyde (MDA), and thiol. Real-time polymerase chain reaction (PCR) was used to assess the expression of the FRMD3 and BMP7 genes in the kidney samples. RESULTS: Diabetes induction increased serum levels of glucose, Cr, urea, MDA, and thiol, but decreased BMP7 and FRMD3 genes expression. Treatment with crocin and losartan decreased these biochemical parameters and increased the expression of the BMP7 and FRMD3 genes. DISCUSSION: Crocin may be a promising therapeutic agent for preventing and improving diabetes-related kidney disease due to its antidiabetic and antioxidant properties.

ABSTRACT

N-Acetylcysteine (NAC) is a chemical compound with anti-inflammatory and antioxidant activity and acts as a free radical scavenger. Elaeagnus angustifolia (EA) is a plant native to the western part of Iran, with antioxidant and anti-inflammatory properties. The present study been taken evaluated the protective effect afforded by EA and NAC extracts on carrageenan-induced acute lung injury in Wistar rats. In this study, 42 rats were randomly assigned into seven groups. NAC and EA extracts were orally administered once/day for 21 continuous days. Pulmonary damage was induced by intratracheal injection of 100 µl of 2% λ-Carrageenan on day 21. Twenty-four hours post-surgery, the rats were euthanized and the samples were collected. Pretreatment with NAC and EA extracts reduced the total and differential cell accumulation as well as IL-6, and TNF-α cytokines. Antioxidant indicators demonstrate that in the groups receiving NAC and EA extract, MDA decreased while thiol and antioxidant capacity elevated. Treatment with NAC and EA significantly reduced Carrageenan-induced pathological pulmonary tissue injury. NAC and EA extract has protective effects on acute carrageenan-induced lung injury.

ABSTRACT

There is an increasing concern over the environmental degradation caused by agricultural activities especially in developing countries which mostly linked to farmer's behavior. Thus, this study aimed to model the responsible attitude and behavior of Iranian farmers in respect to environment. The Theory of Planned Behavior (TPB) was used as a basis for identification of the main determinants of the farmers' responsible environmental behavior. A survey of 400 farmers in the Hamedan Province, selected through a multistage stratified random sampling method. Data was gathered using a questionnaire which its validity and reliability were confirmed by a panel of agricultural and environmental experts and calculated Cronbach's alpha (0.65 ≤ α ≤ 0.80), respectively. Data were analyzed using structural equation modeling (SEM) to evaluate the strength of relationship between the constructs and test the overall model fit. Results of study showed that "using animal manure in the farms" was the most important behavior which farmers are responsibly doing to respect their environment but their least attention is given to "disposing cans and bottles after spraying." Also, the fitness indices of the model revealed that the TPB partially supported the farmers' responsible environmental behavior as perceived behavioral control and environmental attitude positively influenced the farmers' responsible environmental behavior but there was not observed any significant influence by subjective norm. Among the additional incorporated constructs, environmental awareness, ethical commitment, and environmental concern significantly had a positive influence on farmers' behavior by mediating role of the environmental attitude. The inclusion of new constructs in the TPB model was supported through improving the predictive power of the modified model in predicting farmers' responsible environmental behavior.

ABSTRACT

The main purpose of this study was to analyze the perceived outcomes of Good Agricultural Practices (GAPs) technologies adoption in order to sustain citrus farms in Mazandaran province, Iran. Study population consisted of all citrus growers in the villages of 12 counties of Mazandaran province, which a sample of 290 orchardmen were selected through a proportional random sampling technique. A questionnaire was designed to collect data which was both valid and reliable according to expert opinion and Cronbach's alpha coefficient respectively. The results of the factor analysis showed that "market access and safe product exports," "consumer' health and environment-friendly behavior," "safe production and public demand," and "information sharing and strengthening local associations" were the four perceived outcomes of GAPs technologies adoption in citrus farms of Iran. These factors explained 65.02% of the total variance. These four perceived outputs of GAPs support economic, environmental, and social sustainability dimensions respectively.

ABSTRACT

Understanding how to source agricultural raw materials sustainably is challenging in today's globalized food system given the variety of issues to be considered and the multitude of suggested indicators for representing these issues. Furthermore, stakeholders in the global food system both impact these issues and are themselves vulnerable to these issues, an important duality that is often implied but not explicitly described. The attention given to these issues and conceptual frameworks varies greatly--depending largely on the stakeholder perspective--as does the set of indicators developed to measure them. To better structure these complex relationships and assess any gaps, we collate a comprehensive list of sustainability issues and a database of sustainability indicators to represent them. To assure a breadth of inclusion, the issues are pulled from the following three perspectives: major global sustainability assessments, sustainability communications from global food companies, and conceptual frameworks of sustainable livelihoods from academic publications. These terms are integrated across perspectives using a common vocabulary, classified by their relevance to impacts and vulnerabilities, and categorized into groups by economic, environmental, physical, human, social, and political characteristics. These issues are then associated with over 2,000 sustainability indicators gathered from existing sources. A gap analysis is then performed to determine if particular issues and issue groups are over or underrepresented. This process results in 44 "integrated" issues--24 impact issues and 36 vulnerability issues--that are composed of 318 "component" issues. The gap analysis shows that although every integrated issue is mentioned at least 40% of the time across perspectives, no issue is mentioned more than 70% of the time. A few issues infrequently mentioned across perspectives also have relatively few indicators available to fully represent them. Issues in the impact framework generally have fewer gaps than those in the vulnerability framework.
